This diploma thesis describes the design of the illumination system for a Coherence--Controlled Holographic Microscope (CCHM). The theoretical part mentions the history of microscopy, the principle of holography and individual types of interference microscopy. To get closer to the topic, individual light sources and an overview of current illumination systems are mentioned. The diploma thesis also describes the procedure of designing a fiber-optic illumination module for microscopy from optical design through design of construction to the last step, which is assembly and testing of the module.
The effect of manufacturing inaccuracies on the final light intensity distribution of an automotive headlamp

The thesis deals with the issue of headlamp defects arising during production and their impact on the resulting light trail. The thesis describes and analyses defects arising during individual correction loops. Their analysis was carried out in order to provide measures for other projects. The main defects that have been assessed include the position of the light source, the quality of the optical surfaces and the associated quality of the plating, inaccuracies of the optical surfaces, dimensional variations due to installation technology and defects on the cover glass. The proposed measures concern the pressing tool, accuracy of machining and surface polishing. The thesis also includes designing solutions for quick identification of the surface quality of the cover glass by remote projection. Individual defects were described by their originators, their seriousness and their reparability. Based on this information, recommendations for production are proposed.
Combining the light of two sources in the integration sphere

For the development of the new generation of the controlled holographic microscope, is necessary to design a new fluorescence module. The work discusses the basic principles and application of fluorescence microscopy, including a description of the holographic microscope. Searching for available light sources used in fluorescence microscopy is performed and evaluated. The designed source combines radiation with selected excitation wavelengths using an integrating sphere, which also has a homogenizing effect. This innovative design ensures a quick and efficient change of excitation wavelengths due to the fluorophore used and the possibility of a direct connection of the source to the microscope. In order to choose a suitable arrangement for the source, three conceptual designs of its function are proposed. The selected designs were verified by simulation and suitable optical components were selected. The resulting construction is tested.
Homogenized illumination source for microscopy

This thesis deals with the design of an illumination system for homogeneous illumination of the sample of a coherence-controlled holographic microscope. The theoretical part of the thesis briefly describes the principle of interference and holographic microscopy in connection with the use of a suitable illumination system for microscopy, including methods of light homogenization. The definition of the parameters of the new miniaturized source is based on this part. The work includes two designs of an illumination system with different use of microlens arrays. Optical simulations are performed, which verify the correct selection of all optical elements of the illumination system. Following the optical models, optomechanical designs are created. These are compiled and tested. The light efficiency of the sources and the distribution of light intensity values in their output planes are measured.
